In reptiles,administrationofopioiddrugshasyieldedunexpectedresultswithrespecttoanalgesia. Tapentadol(TAP)isanovelatypicalopioiddruglabelledforhumanuse.Theobjectiveofthisstudywas to evaluatethepharmacokineticsandthepharmacodynamicsofthisdruginyellow-belliedsliders,aftera single intramuscular(IM)injectionof5mg/kgofTAP.Turtles(n ¼ 9) wererandomlyassignedto2 treatment groups,accordingtoasingle-dose,single-treatment,unpaired,2-periodcrossoverdesign. Group A(n ¼ 5) receivedasingleIM(proximalfrontlimb)doseofTAP(5mg/mL)at5mg/kg.GroupB (n ¼ 4) receivedasingleIMinjectionofsaline(equivalentvolumetoopioidvolumes)ofTAPatthesame site. Aftera1-monthwashoutperiod,groupswererotatedandtheexperimentrepeated.TAPplasma concentrationsweredeterminedbyavalidatedhigh-performanceliquidchromatography-fluorescence method, andaninfraredthermalstimuliwasappliedtotheplantarsurfaceoftheturtles’ hind limbsto evaluatethethermalwithdrawallatency(TWL).TAPplasmaconcentrationsweredetectablebetween1 and 24hour(s)(1619and37ng/mL,respectively).TheTAP-treatedgroupshowedanincreaseinTWL1 hour afterdrugadministration(13.32 6.40 seconds).Subsequently,TWLdecreasedwithtimeand significant differencesbetweentreatmentandcontrolgroupswereapparentupto10hoursfollowing treatment. Alinearrelationship(r2 ¼ 0.99)betweenTAPplasmaconcentrationandeffectwasfound. Given these findings,TAPappearstobeanattractiveoptionforantinociceptioninturtles,owingtoits rapid onsetandacceptabledurationofeffect.
