Red deer(Cervus elaphus) haveapronouncedseasonalityintheirphysiology.Theaimof this studywastoassesstheeffectoftheseasononreddeerresponsivenesstoskintesting with thephytohaemagglutinin(PHA)mitogen.Studysubjectsincluded270farmedadult red deer(19stagsand251hinds).TheskintestingwascarriedoutbetweenJanuary2009 and August2010.Theanimalswereinjectedintradermallywitha0.1mlvolumecontaining 250g ofPHAdilutedinphosphatebufferedsaline.Theskinfoldthicknesswasmeasured immediately priortoinjectionand72hafteradministration,alwaysbythesamepersonand with threerepeatspermeasurement.Singleeffectsofsexandtimeonskintestresponsive- ness weresignificant(p < 0.001)aswellastheirinteraction(p < 0.001).Inwinter(January), and consideringtheaverageoftwoyears,theskinfoldincreaseinresponsetotheintra- dermal injectionof250g PHAwas2.1timeslargerinstagsand1.4timesinhindsthan in summer(August).Whilestagshad1.3timeslargerresponsesthanhindsinwinter,the inverse occurredinsummer,with1.1timeslargerresponsesinhinds.Wealsoevidenced a limitedinter-annualvariationofskinfoldincreaseinresponsetoPHAinreddeer.These findings haveimportantconsequencesregardingtheinterpretationofskintestresultsin the ante-mortemdiagnosisoftuberculosisandparatuberculosis,atleastindeer.
