Objectives: The aim of the trial was to screen out lymph nodes from the carcass of pigs fed an experimental diet based on ripe unprocessed acorns by means of natural polyphenols as biological markers. Therefore, a method to evaluate lymphnodes (lnn) pigmentation from carcasses of pigs from a traditional feeding practice based on acorns offer was tested. Methods: A total of 20 cross-bred pigs were divided into 2 experimental groups: n= 10 pigs (traditional group, T) were fed an experimental diet based on a traditional recipe (acorns: 50%, wheat and faba beans based), where polyphenolic compounds from acorns supply was 7.80% in DM of the acorns based diet; n= 10 (control group, C) received a conventional complete diet for pigs. Pigs (T/C) were fed for 4 weeks. Animals were slaughtered and lnn. from different anatomical districts inspected. Histological conventional methods (HE stain) were used and slides were observed at light microscope. At confocal microscope auto-fluorescence was checked. Results: A macroscopic pigmentation of lnn. occurred in both groups, but C lnn. pigmentation (dark red) appeared randomly and only partly in some lnn. (renal or sub-iliac). T lnn from showed a dark brown staining diffused to every whole lnn homogeneously, associated with a brownish colour of muscles. A stronger pigmentation involved left iliac and sub-iliac lnn. in both groups. At light microscope intracytoplasmic granules were found in macrophages from both groups, but, at confocal an intense fluorescence was observed in medial-iliac lnn from T carcasses (green emission) Conclusions: Our results don’t seem to confirm the marked evidence reported in reference about lnn pigmentation in order to univocally discriminate carcasses of pigs fed acorns. Anyway, the intense green fluorescence of medial iliac lnn. is worthy to be investigated to set up a proper method to discriminate carcasses of swine on the basis of polyphenols auto-fluorescence.
